    Adaptive object management for distributed systems

    This thesis describes an architecture supporting the management of pluggable software components and evaluates it against the requirement for an enterprise integration platform for the manufacturing and petrochemical industries. In a distributed environment, we need mechanisms to manage objects and their interactions. At the least, we must be able to create objects in different processes on different nodes; we must be able to link them together so that they can pass messages to each other across the network; and we must deliver their messages in a timely and reliable manner. Object based environments which support these services already exist, for example ANSAware(ANSA, 1989), DEC's Objectbroker(ACA,1992), Iona's Orbix(Orbix,1994)Yet such environments provide limited support for composing applications from pluggable components. Pluggability is the ability to install and configure a component into an environment dynamically when the component is used, without specifying static dependencies between components when they are produced. Pluggability is supported to a degree by dynamic binding. Components may be programmed to import references to other components and to explore their interfaces at runtime, without using static type dependencies. Yet thus overloads the component with the responsibility to explore bindings. What is still generally missing is an efficient general-purpose binding model for managing bindings between independently produced components. In addition, existing environments provide no clear strategy for dealing with fine grained objects. The overhead of runtime binding and remote messaging will severely reduce performance where there are a lot of objects with complex patterns of interaction. We need an adaptive approach to managing configurations of pluggable components according to the needs and constraints of the environment. Management is made difficult by embedding bindings in component implementations and by relying on strong typing as the only means of verifying and validating bindings. To solve these problems we have built a set of configuration tools on top of an existing distributed support environment. Specification tools facilitate the construction of independent pluggable components. Visual composition tools facilitate the configuration of components into applications and the verification of composite behaviours. A configuration model is constructed which maintains the environmental state. Adaptive management is made possible by changing the management policy according to this state. Such policy changes affect the location of objects, their bindings, and the choice of messaging system

    LASER SAFETY FOR THE EXPERIMENTAL HALLS AT SLAC’S LINAC COHERENT LIGHT SOURCE (LCLS) *

    The LCLS at the SLAC National Accelerator Laboratory will be the world’s first source of an intense hard x-ray laser beam, generating x-rays with wavelengths of 1nm and pulse durations less than 100fs. The ultrafast x-ray pulses will be used in pump-probe experiments to take stop-motion pictures of atoms and molecules in motion, with pulses powerful enough to take diffraction images of single molecules, enabling scientists to elucidate fundamental processes of chemistry and biology. Ultrafast conventional lasers will be used as the pump. In 2009, LCLS will deliver beam to the Atomic Molecular and Optical (AMO) Experiment, located in one of 3 x-ray Hutches in the Near Experimental Hall (NEH). The NEH includes a centralized Laser Hall, containing up to three Class 4 laser systems, three x-ray Hutches for experiments and vacuum transport tubes for delivering laser beams to the Hutches. The main components of the NEH laser systems are a Ti:sapphire oscillator, a regen amplifier, green pump lasers for the oscillator and regen, a pulse compressor and a harmonics conversion unit.     A review of issues in seagrass seed dormancy and germination: implications for conservation and restoration

    Seagrasses have received considerable attention over the past 2 decades because of the multiple ecological roles they play in estuarine and coastal ecosystems and concerns over worldwide losses of seagrass habitat due to direct and indirect human impacts. Restoration and conservation efforts are underway in some areas of the world, but progress may be limited by the paucity of information on the role of seeds in bed dynamics. Although flowering occurs in most of the 58 seagrass species, seed germination data exist for only 19 of the 42 species that have some period of dormancy, with only 93 published references to field and/or laboratory studies. This review addresses critical issues in conservation and restoration of seagrasses involving seed dormancy (e.g. environmental vs physiological), existence and type of seed bank (transient or persistent), and factors influencing seed germination (e.g, salinity, temperature, light). Results of many earlier published studies relating seed germination to various environmental factors may need re-examination given more recent published data which show a confounding influence of oxygen level on the germination process. We highlight the importance of conducting ecologically meaningful germination studies, including germination experiments conducted in sediments. We also identify questions for future research that may figure prominently in landscape level questions regarding protected marine or estuarine reserves, habitat fragmentation, and restoration

    Genetic Variability Overrides the Impact of Parental Cell Type and Determines iPSC Differentiation Potential

    Reports on the retention of somatic cell memory in induced pluripotent stem cells (iPSCs) have complicated the selection of the optimal cell type for the generation of iPSC biobanks. To address this issue we compared transcriptomic, epigenetic, and differentiation propensities of genetically matched human iPSCs derived from fibroblasts and blood, two tissues of the most practical relevance for biobanking. Our results show that iPSC lines derived from the same donor are highly similar to each other. However, genetic variation imparts a donor-specific expression and methylation profile in reprogrammed cells that leads to variable functional capacities of iPSC lines. Our results suggest that integration-free, bona fide iPSC lines from fibroblasts and blood can be combined in repositories to form biobanks. Due to the impact of genetic variation on iPSC differentiation, biobanks should contain cells from large numbers of donors.Peer reviewe

    Unresolved issues with the assessment of multidecadal global land surface temperature trends

    This paper documents various unresolved issues in using surface temperature trends as a metric for assessing global and regional climate change. A series of examples ranging from errors caused by temperature measurements at a monitoring station to the undocumented biases in the regionally and globally averaged time series are provided. The issues are poorly understood or documented and relate to micrometeorological impacts due to warm bias in nighttime minimum temperatures, poor siting of the instrumentation, effect of winds as well as surface atmospheric water vapor content on temperature trends, the quantification of uncertainties in the homogenization of surface temperature data, and the influence of land use/land cover (LULC) change on surface temperature trends. Because of the issues presented in this paper related to the analysis of multidecadal surface temperature we recommend that greater, more complete documentation and quantification of these issues be required for all observation stations that are intended to be used in such assessments. This is necessary for confidence in the actual observations of surface temperature variability and long-term trends

The objective of this work is to describe how individual nurses and midwives, organisations and education providers could use the newly developed National Nursing and Midwifery Digital Health Capability Framework to build digital health capability.     Reversing age: Dual species measurement of epigenetic age with a single clock

    Young blood plasma is known to confer beneficial effects on various organs in mice. However, it was not known whether young plasma rejuvenates cells and tissues at the epigenetic level; whether it alters the epigenetic clock, which is a highly-accurate molecular biomarker of aging. To address this question, we developed and validated six different epigenetic clocks for rat tissues that are based on DNA methylation values derived from n=593 tissue samples. As indicated by their respective names, the rat pan-tissue clock can be applied to DNA methylation profiles from all rat tissues, while the rat brain-, liver-, and blood clocks apply to the corresponding tissue types. We also developed two epigenetic clocks that apply to both human and rat tissues by adding n=850 human tissue samples to the training data. We employed these six clocks to investigate the rejuvenation effects of a plasma fraction treatment in different rat tissues. The treatment more than halved the epigenetic ages of blood, heart, and liver tissue. A less pronounced, but statistically significant, rejuvenation effect could be observed in the hypothalamus. The treatment was accompanied by progressive improvement in the function of these organs as ascertained through numerous biochemical/physiological biomarkers and behavioral responses to assess cognitive functions. Cellular senescence, which is not associated with epigenetic aging, was also considerably reduced in vital organs. Overall, this study demonstrates that a plasma-derived treatment markedly reverses aging according to epigenetic clocks and benchmark biomarkers of aging.Fil: Horvath, Steve.
